The nearest railway station from Mumbai is Neral. From Neral you can take Toy Train towards Matheran.

From Pune :

Catch Mumbai bound train and get off at Karjat, and need to catch the train going towards CST and get down at Neral station. From Neral you can take Toy Train towards Matheran.
