Car, Bus and Train - all options are there. Distance is around 375 km. Train would take close to 12 hrs.

There is a daily train from Mathura to jaipur. after reaching Jaipur take a train for Ajmer or can go by bus and from Ajmer can take a bus easily to Pushkar. OR you can take a direct train from Jaipur to Pushkar as well.
