*The route you mensioned are totally opposite and from different different states , Mussoorie and auli are from uttrakhand and kufri ,Shimla ,Rohtang are from Himachal Pradesh .
*you can do only once at a time .whether you can explore uttrakhand by Mussoorie to auli route covering kedarkantha, joshimath , Rishikesh or you can travel to himachal-pradesh by this route ,delhi Shimla-kufri-manali-rohtang-dalhausie-dharmashala and back .
*since you mensioned that you are planning for bike trip ,I would suggest you to better explore Himachal this is because due to early infrastructural development,it has better roadways and transportation facilities than uttrakhand .

If you have your own bike then you can start from Dehradun but if you want to rent a bike then firstly you should go Rishikesh from there you can take bike on rent and they have so much of variety, choose which one you want then starts your journey
Get down at Himachal Pradesh station, take a bus to Kasol, explore the valley, Trek to chalal, join a camp there, live a night, return back to Kasol, take a state transport back to Himachal station.
